Reliable defender Vlastuin re-joins the line-up after a back complaint saw him miss just his second game of the season last weekend.
Important midfielder and 2023 Jack Dyer Medallist Taranto returns to the side after being unavailable for Round 19 due to concussion.
"Tim bounced out (of concussion protocols) straight away and trained really well on Tuesday," Senior coach Adem Yze said when speaking to the media on Thursday morning.
"Nick had a grumbly back for around a month. He's one of our leaders who just kept pushing through for four weeks, and we needed to break the cycle.
"He's bounced out of that really well."
The two inclusions replace captain Toby Nankervis, who suffered a concussion last Saturday night against Port Adelaide, and Tyler Sonsie, who has been omitted.
Richmond’s three emergencies are Jacob Bauer, Matt Coulthard and Tyler Sonsie.
Talented tall Noah Balta will play his 100th AFL game on Sunday.
"This season, he's grown. His leadership's grown, he's moved into our performance group which is our emerging leaders," Yze said of Balta.
"He's just a quality contest player... We love having him around and we can't wait to see what he can do this weekend."
Yze said it was important for the Tigers to finish the home and away season strongly.
"We want to play a brand of footy that can stand against a really good team," Yze said.
"Last week was some quality footy interstate against a really good team for three quarters. We just want to maintain that."
The clash at the 'G kicks off at 1:10pm (AEST) and can be viewed live on Fox Footy and Kayo Sports.
