A third-quarter burst from the Tigers proved the difference between the two teams, in a scrappy affair in front of 52,564 on Friday night footy.
Jack Riewoldt was the leading goalkicker on the ground with three, while Alex Rance was again strong in defence.
Rance was the dominant player on the field in the first half with 18 disposals, and he finished with 22, to go with eight marks.
Shaun Grigg was a terrific four-quarter contributor, finishing with 25 disposals and six tackles, while captain Trent Cotchin overcame a slow start to win 23 possessions and kick two important goals.
In his first game of the season, Shaun Hampson was a solid contributor, dominating the hitouts with 34, and taking eight marks.
The Tigers made the perfect start to the match, when Riewoldt pounced on a loose ball and kicked truly inside the first minute.
The next goal came more than 20 minutes later, when Alex Rance found Brett Deledio with a pin-point pass inside 50.
In between, Carlton had the majority of the genuine goal-scoring opportunities, but failed to capitalise.
The Blues could only muster one goal for the quarter, a set shot bomb from inside the centre square by Levi Casboult.
Richmond drew first blood in the second quarter, when Shane Edwards swooped on a loose ball on the boundary and ran into an open goal.
Carlton answered though, with a fast break ending with a running goal from Andrejs Everitt.
Approaching half time, Cotchin, who had 10 disposals in the term, capitalised on a 50-metre penalty to extend the margin to 10 points, making amends for a set shot miss a few minutes earlier.
Richmond made its move in the third quarter, booting the first four goals to set up a match-winning lead.
Shaun Grigg set the wheels in motion with a long-range set shot, before Cotchin kicked his second, snapping from the pocket after strong forward pressure from Grigg and Riewoldt.
Riewoldt then kicked two goals in quick succession to extend the margin to 35 points.
Carlton rallied briefly, kicking two quick goals heading into three quarter time, but a goal to Ty Vickery at the seven minute mark of the final term secured the four points for the Tigers.
With a 9-5 win-loss record after 15 rounds, Richmond takes on St Kilda at Etihad Stadium next Sunday afternoon.
